  • PharmacologyA fibrate. It is a lipid lowering agent used for controlling the high cholesterol and triacylglyceride level in the blood. It increases lipoprotein lipase activity to promote the conversion of VLDL to LDL, and hence reduce the level of VLDL. It is proved that it can increase the level of HDL as well.
  • Absorption_value96.0
  • Absorption (description)Readily and almost completely absorbed after oral administration. Between 95% and 99% of an oral dose of clofibrate is excreted in the urine as free and conjugated clofibric acid; thus, the absorption of clofibrate is virtually complete.

  • MetabollsmRapidly hydrolysed by serum enzymes to clofibric acid, 2-(4-chlorophenoxy)-2-methylpropionic acid (active), which is conjugated with glucuronic acid.
  • Half lifeclofibric acid 12~25 h but may be decreased when free fatty acid concentrations are high; longer half-lives have been reported after repeated doses, and in subjects with renal impairment.
  • ExcretionAbout 50 to 85% of a dose is excreted in the urine in 48 h, mostly as conjugated clofibric acid.
  • Urinary ExcretionN/A
  • CleranceN/A
  • ToxicityA 15-year-old boy who took 24.5 g of clofibrate in a suicide attempt complained of headache, pain in the arms, and inability to walk, but no evidence of toxicity was found in the following 5 days. [A. H. Greenhouse,JAMA,1968, 204, 402–403.]
